Oxidative Modifikationen oder Destruktionen sind die Ausgangsbasis zahlreicher Erkrankungen, wie z. B. entzündlicher Prozesse, Krebs, neurodegenarativer Prozesse, Katarakt und Atherosklerose.
Atherosklerotische Gefäßläsionen und ihre Folgen sind die häufigste Todesursache weltweit und daher von enormer gesundheitspolitischer Bedeutung. Besonders betroffen sind Patienten mit Diabetes mellitus, da sie bewiesenermaßen ein stark erhöhtes Risiko für Atherosklerose haben.
In dieser Arbeit wird ein Beitrag zum Verständnis des erhöhten Risikos von Diabetikern für atherosklerotische Gefäßläsionen geleistet, sowie die Wirkung von synthetischen Antioxidantien wie "Pangamsäure" auf den initialen Prozess der Atherosklerose, die low density Lipoprotein (LDL) Oxidation, untersucht. Weiterhin wird die Cooperativität von natürlichen Antioxidantien wie Coenzym Q10, a-Tocopherol und Dihydroliponsäure beim Schutz des LDL vor Oxidation beschrieben.
Die "Pangamsäure" ist eine Substanz, die seit den 50er Jahren in der Literatur immer wieder auftaucht und der neben zahlreichen anderen Eigenschaften vor allem eine Stimmulation des Sauerstoffmetabolismus nachgesagt wird. In dieser Arbeit wurde zum ersten mal eine vollständige Strukturanalyse der käuflich erhältlichen und in einigen pharmazeutischen Präparaten enthaltenen "Pangamsäure" durchgeführt und ihr antioxidatives Potential sowie der Einfluß auf die oxidative Respiration untersucht.
[The fact that the consequences of atherosclerosis are the most frequent cause of death explains the enormous meaning of this toping in regard to public health. Especially diabetes mellitus patients are concerned since their enhanced atherosclerosis risk is well known. This thesis makes a contribution to a better comprehension of the higher risk of diabetes patients. Futhermore the effect of supposed antioxidants such as coenzyme Q10 and "pangamic acid (vitamin B15)" in the low density lipoprotein oxidation which is the initial step in atherosclerosis has been investigated. Among many other properties "pangamic acid" is supposed to stimulate the oxygen metabolism. In this thesis the very first time a complete structure analysis of "pangamic acid" has been carried out including the examination of its antioxidative potential and its impact on the electron transport of isolated mitochonria.]
